Antioxidant Activity of Ethyl acetate Fraction of the Guzeunggupo-procossed Eucommia ulmoides Oliver in Caenorhabditis elegans
In the course of finding antioxidants in natural products, it was found that the ethyl acetate (EA) soluble fraction of methanol extract and its fractions of Eucommia ulmoides Oliver barks showed the strong DPPH radical scavenging activities. The guzeunggupo-processed E. ulmoides barks extract and its fractions revealed the more potent activities. Among the fractions, the EA fraction of the guzeunggupo-processed E. ulmoides (EAGE) barks showed the best DPPH radical scavenging activity. The EAGE barks was measured for the antioxidant related activities like catalase, superoxide dismutase (SOD), oxidative stress tolerance and reactive oxygen species (ROS) level by using C. elegans along system. Furthermore, to confirm the relationship of stress response genes derived from increased stress resistance of EAGE-treated C. elegans, the CF1553 transgenic strain was used to confirm SOD-3 expression. As a results, EAGE enhanced the SOD and catalase activities, and decreased ROS levels in a dose-dependent manner. And, the EAGE-treated CF1553 worm revealed higher SOD-3::GFP intensity than the control worms.
