“Method Development And Validation Of Simultaneous Estimation Of Olmesartan & Hydrochlorothiazide In Bulk & Formulation Byrp- Hplc Method”

A simple fast precise reverse phase isocratic high performance liquid chromatographic (HPLC) method has been developed for simultaneous estimation of Olmesartan & Hydrochlorothiazide in bulk and formulation by RP-HPLC method. The chemical name of Olmesartan is 5-(2-hydroxypropan-2-yl)-2-propyl-3-[[4-[2-(2H-tetrazol-5- yl)phenyl]phenyl]methyl]imidazole-4-carboxylic acid. The method was validated for several parameters such as accuracy precision robustness as per ICH guidelines. The RP-HPLC method for the estimation of Olmesartan and Hydrochlorthiazide in bulk and pharmaceutical dosage formwas developed. The quantification was carried out by using ZobraxSB-Aq column (250 mm × 4.6 mm, 5 m) as stationary phase,0.1% Orthophosphoricacid:Acetonitrile [60:40] as mobile phase. Mobile phase was maintained at a flow rate of 1.0ml/min at 256nm. The drugsOlmesartan and Hydrochlorthiazide eluted at 4.93 and 3.63 minutes. Validation data demonstrates that, this method is accurate, precise, simple, economic and can be used in the routine analysis of Olmesartan and Hydrochlorthiazide in various formulations. Different mobile phase combination of acetonitrile and 0.1% orthophosphoric acid were tried for better resolution of the chromatogram.
