An In Silico Method For Design Of Soluble Fusion Partner For Expression Of Modified Glucagon-Like Peptide-1 In E.coli BL21(DE3).
Glucagon-like peptide-1 (GLP-1) is an essential therapeutic agent for diabetes and obesity; however, its poor solubility presents significant challenges in production, leading to complex purification steps, increased processing time, and higher costs. This study introduces a novel strategy employing an in-silico approach to identify a soluble fusion partner (sfp) to enhance mGLP-1 solubility.Using Protein-sol tool, we identified a fusion partner that can be fused to mGLP-1 via a His6-EK linker resulted a solubility score of 0.947.The resulting fusion construct was cloned into the pET22b vector and expressed in Escherichia coli.Expression analysis revealed that the fusion partner effectively enhanced GLP-1 solubility, maintaining it across various IPTG concentrations. This approach offers a promising solution to simplify GLP-1 protein expression, potentially reducing purification complexity and manufacturing costs. These In-Silico method could also be applied for expression of other proteins and peptides.
