Assessment of Bioavailability Enhancement from TransdermalFormulations of Buspirone hydrochloride and Trifluoperazinehydrochloride using Rabbit as Animal Model
This study involved pharmacokinetic analysis of transdermal formulations of two model psychotropic drug candidates: buspirone hydrochloride and trifluoperazine hydrochloride and its comparison with oral dosing.A transdermal patch of selected drugs were developed using chitosan and PVA as polymers. The in-vivo pharmacokinetic studies were performed in rabbits by oral and transdermal application of drugs and various pharmacokinetic parameters like Tmax, Cmax and AUC were determined. The skin irritation study of the patch was also carried out to establish the dermal safety of the formulations. The pharmacokinetic studies confirmed significant enhancement in the bioavailability of transdermal formulations of buspirone HCl and trifluoperazine HCl compared to the oral solution. The results presented statistical significance at 95% confidence (P<0.05).Dermal irritation study revealed no significant irritation or edema upon removal of the transdermal patch in rabbits, suggesting that the formulation is well-tolerated and less likely to induce adverse effects.A stable plasma drug profile was maintained up to 96 hrs after transdermal dosing of the selected drugs.
