The Role Of Adipocyte Factor Irisin In The Progression And Prevention Of Breast Cancer
PURPOSE: To explore the inhibitory effect of exercise factor irisin on obesity and breast cancer, and to provide theoretical support for the development of related antitumor drugs. METHODS: Mature adipocytes and human breast cancer cell line MDA-MB-231 cells were co-cultured by Transwell culture system and divided into four groups. Blank control group (MDA-MB-231 cells alone, that is, control group), mature adipocyte co-culture group (MDA-MB-231+ mature adipocytes, Adipocytes), MDA-MB-231 cell culture +Irisin group (Irisin group), mature Adipocytes coculture +Irisin group (MDA-MB-231+ mature Adipocytes +Irisin group). The changes of invasion and migration ability and inflammatory factors of breast cancer cells before and after co-culture were measured. RESULTS: The results of inter-group covariance showed that the migration ability of irisin group was significantly inhibited compared with the control group after co-culture with Irisin treatment (71 ± 8, P < 0.05), the migration ability of Adipocytes group and compared group was significantly inhibited (141 ± 8, P < 0.05); Compared with the control group, the invasion ability of MDA-MB-231 cell culture plus Irisin group was significantly inhibited (1.427 ± 0.09, <0.05), the migration ability of Adipocytes and Adipocytes+Irisin groups was significantly inhibited (1.770 ± 0.043,) compared with that of Adipocytes+Irisin groups. In terms of inflammatory factors, compared with the control group, after intervention, IL-6 in Irisin group (10.3011 ± 0.1003, P < 0.05), TNF-α(203.3827 ± 2.454,P < 0.05), Leptin (6.6874 ± 0.0895, P < 0.05); IL-6 (11.5456 ± 0.188, P < 0.05) in Adipocytes+Irisin groups;TNF-α(213.94 ± 0.82, P < 0.05) Leptin (7.8212 ± 0.1101, P < 0.05) were improved. CONCLUSIONS: Exercise factor irisin can significantly inhibit the invasion and migration of breast cancer cells, reduce the content of inflammatory factors, and improve the health of breast cancer patients.
