Anti-skin Aging Effects Of Aerobic And Resistance Exercise Via IL-18/IL-18R Pathway In Samp8 Mice
Aging is a chronic inflammatory process that causes changes such as skin thinning, fragility, hair loss, hyperpigmentation, and delayed wound healing. As a therapeutic target for inflammatory diseases, IL-18 has strong pro-inflammatory activity. The IL-18/IL-18R pathway may be an important pathway against skin aging. Exercise is a key strategy to combat aging. Previous studies have shown that exercise can reduce the expression level of IL-18 in the body, but its action pathway and regulatory mechanism need to be further clarified. PURPOSE: To investigate the effects of aerobic and anti-resistance exercise on skin aging in SAMP8 mice via IL-18/IL-18R pathway, and provide new ideas for anti-skin aging and prevention and treatment of chronic inflammatory skin-related diseases. METHODS: Twenty-four healthy male 28-week-old rapidly aging SAMP8 mice were randomly divided into a model group (Group M), an aerobic exercise group (Group E), and a resistance exercise group (Group R), with 8 mice in each group, and 8 same-aged fast-aging SAMR1 mice as a control group (Group C). Group E was trained with running table training at a constant speed (1 time per day, 5 times per week) for 8 weeks, and Group M was trained with incremental weight-bearing ladder training for 8 weeks (1 time every other day, 3 times per week). (1) ELISA to detect the contents of IL-18, IL18R and IL18BP; (2) Immunofluorescence to double-stain IL18 and IL18R; (3) RT-qPCR to detect the contents of IL-18, IL-18R gene expression level. RESULTS: Compared with the group C, the mice in the group M showed skin IL-18 and IL-18R content was significantly higher, and IL-18BP content was significantly lower(P < 0.05 or 0.01); co-localization phenomenon existed between IL18 and IL18R; and skin IL-18 and IL18R mRNA expression were significantly higher(P < 0.01). Compared with the group M , skin IL-18 and IL-18R contents were significantly reduced in both group E and R(P < 0.01), IL-18BP contents were significantly increased(P < 0.01), and co-localized expression of IL-18 and IL-18R were significantly reduced; and mRNA expression of IL-18 and IL-18R was significantly reduced in both group E and R (P < 0.01). CONCLUSION: Both aerobic exercise and resistance exercise may delay age-related skin aging in SAMP8 mice by down-regulating the IL-18/IL-18R pathway.
