Exploring the Molecular Impact of Cancer-associated Histone Mutations on Nucleosome Stability, Interactions and Higher-Order Structures
Nucleosomes, as key players in epigenetic regulation, interact with various chromatin factors and enzymes. Recently, a variety of studies have highlighted the significant roles that histone mutations play in the progression of various cancers. Cancer-associated mutations can disrupt nucleosome stability, dynamics and interactions, leading to profound effects on higher-order chromatin structure and epigenetic processes. In this review, we summarize the latest research that provides a comprehensive analysis of how histone mutations in cancers influence nucleosome dynamics, stability and interactions by altering their physicochemical properties. Moreover, we briefly explore the molecular mechanisms by which these mutations disrupt higher-order chromatin organization and epigenetic landscape.
