Spectral Analysis of Novel Minocycline/Zn Complex with Promising Anticancer Activities Against Large Lung Cancer Cells (H460), Antibacterial and Antioxidant Activities Against Acrylamide-Induced Pulmonary Toxicity in Male Rats
Background and Objective: Minocycline (Mino) is a broad spectrum tetracycline antibiotic, as novel drug complexes attracted attention based on the reduction of possible side effects and producing compounds with new potent capacities, thus the main objective of the study is to assess the potent capacities of new Mino/Zn drug complex. Materials and Methods: Minocycline/zinc (Mino/Zn) novel complex was synthesized and characterized using elemental analysis, spectroscopic methods (IR,1H-NMR, UV and X-ray diffraction), magnetic susceptibility, SEM and TEM. An experimental model was used via 32 male rats, divided into 4 groups: Control, Acy, Mino/Zn and Acy plus Mino/Zn. Inflammation markers (CRP, IL-6 and TNF-α), antioxidant enzymes (CAT, SOD and GPx), lipid peroxidation marker (MDA), histological and caspase-3 variations were all investigated in lung tissues, beside estimation of cytotoxicity against large lung cancer cells (H460) and antibacterial activity. Results: The non-electrolytic nature of Mino/Zn novel complex was confirmed based on value of molar conductance, Mino is coordinated with Zn metal ion through oxygen atoms of (C = O), ketonic and hydroxyl groups. The surface morphology observed via SEM confirmed that Mino/Zn novel complex appeared as small rectangular projections. The TEM showed formation of black spots for Zn/Mino with particles ranging from 7-19 nm. The Mino/Zn induced highly potent anticancer activities by inhibition of H460 proliferation, the viability of cancer cells was recorded 7.98 μg/mL at concentration of 100 ug/mL. The Mino/Zn alleviated Acy pulmonary toxicity and inflammation markers and has potent antibacterial activities against the bacterial strains (Bacillus subtilis, Staphylococcus aureus and Escherichia coli) at very low concentrations. The Mino/Zn can inhibit viral replication and binding to (ACE2) and the main protease inhibitor Mpro receptors via molecular dynamics simulation. Conclusion: Novel Mino/Zn novel complex is an effective antioxidant, anticancer and antibacterial agent with cellular inhibitory effect against SARS-CoV-2 via molecular docking simulation.
